Garcinielliptone FC: antiparasitic activity without cytotoxicity to mammalian cells.

چکیده
Garcinielliptone FC (GFC) is a natural prenylated benzophenone found in the seeds of Platonia insignis Mart. (Clusiaceae), a native Brazilian plant. It has been chemically characterized and it is known that GFC has several biological activities such as antioxidant and vasorelaxant properties. In this study, we report the in vitro effect of GFC against the blood fluke Schistosoma mansoni, the parasite responsible for schistosomiasis mansoni. The anti-S. mansoni activity and cytotoxicity toward mammalian cells were determined for the compound. GFC⩾6.25 μM showed antischistosomal activity and confocal laser scanning microscopy analysis demonstrated several morphological alterations on the tegument of worms, and a correlation between viability and tegumental damage was observed. In addition, at sub-lethal concentrations of GFC (⩽3.125 μM), the number of S. mansoni eggs was reduced. More importantly, GFC exhibited no activity toward mammalian cells and, therefore, there is an appreciable selectivity of this compound against the helminths. In conclusion, these findings indicate the potential of GFC as an antiparasitic agent.

متن کاملIndole alkaloids from Aspidosperma rigidum and A. schultesii and their antiparasitic effects.
Five oxindole alkaloids, three plumerane-type alkaloids, subtype haplophitine, and one aspidospermatane-type alkaloid, subtype tubotaiwine, were isolated from the medicinal plants Aspidosperma rigidum and A. schultesii. One compound was identified as the transoid conformer of 18-oxo-O-methylaspidoalbine which was not previously described. The antiparasitic activity of all compounds against Tryp...

The leishmaniasis and Chagas diseases constitute a serious public health problem worldwide with few and ineffective treatment options. The search for new antiparasitic candidates at the initial steps of drug discovery and development is still necessary. The synthesis of 22 de novo synthetized N,N'-dihetaryl-alkyldiamine derivatives and in vitro antiparasitic activity were evaluated for the firs...
متن کاملArylmethylamino steroids as antiparasitic agents
In search of antiparasitic agents, we here identify arylmethylamino steroids as potent compounds and characterize more than 60 derivatives. The lead compound 1o is fast acting and highly active against intraerythrocytic stages of chloroquine-sensitive and resistant Plasmodium falciparum parasites (IC50 1-5 nM) as well as against gametocytes. In P. berghei-infected mice, oral administration of 1...
متن کاملEffect of Soluble HLA Class I Molecule on NK/LAK Cells Activation Induced by Poly I:C
Background: Natural Killer cells express killer inhibitory receptors specific for HLA-class I molecules. These receptors could induce signals that determine NK cells ability to mediate cytotoxicity. Purified soluble form of HLA class I molecules (sHLA) could bind to NK cell receptors and down-regulate the NK killer function. Objective: To evaluate the influence of sHLA and two monoclonal an...
